With both teams focused on winning silverware, as shown by their recent league performances, managers Ange Postecoglou and Ruben Amorim are likely to field their strongest possible lineups for the match at San Mames.
Ahead of Wednesday's game, we provide the latest team news for both sides, starting with Tottenham.
Son Heung-Min
Son Heung-Min has returned to action, starting in the 2-0 loss to Crystal Palace and the 2-0 defeat at Aston Villa on Friday. With match minutes under his belt, it's expected that the Tottenham captain will start in Spain.
Pape Sarr
Pape Sarr started in the 2-0 loss at Villa Park but was substituted early in the second half due to a back injury. While Ange Postecoglou has downplayed concerns about his availability, the Senegal international is listed as doubtful for Wednesday's match.
Lucas Bergvall
Shortly after signing a new deal, it was announced before the first-leg win over Bodø/Glimt that Lucas Bergvall had sustained an ankle injury. James Maddison

Dejan Kulusevski
While Dejan Kulusevski had been struggling for form after recovering from a foot issue, he was likely to feature in Maddison's absence. However, Kulusevski has undergone patella surgery following an injury in the 2-0 defeat to Crystal Palace and will miss the game.
Radu Dragusin
With the defense fully fit, Radu Dragusin would not have started on Wednesday. Even so, the Romania international has been out since sustaining an ACL injury against Elfsborg in January and remains unavailable.
Dane Scarlett
The win over Elfsborg in January was notable for Dane Scarlett, who came off the bench to provide an assist in the home victory. However, Scarlett is currently absent after undergoing groin surgery earlier this month.
Timo Werner
Timo Werner's time at Tottenham has effectively ended after he suffered a hamstring injury earlier this month. Werner hasn't played since the 1-0 home loss to Manchester City in February and was not included in Tottenham's Europa League squad for the latter part of the season, so he wouldn't have been available anyway.
